Hybridization studies with an antibody heavy chain mRNA.

چکیده
In a previous report we presented evidence which indicated that antibody diversity is generated in large part by somatic processes [I] . That evidence was based on DNA-RNA hybridization studies with a purified mouse a-chain mRNA. The present report deals with similar studies using a preparation of mRNA coding for a mouse heavy chain. The results confirm and extend our previous finding, namely there are a very limited number possibly a single copy per subgroup of genes coding for the variable region of immunoglobulin polypeptides.

Localization of cardiac (alpha)-myosin heavy chain mRNA is regulated by its 3' untranslated region via mechanical activity and translational block.
We have altered the spontaneous contractile activity of neonatal cardiac myocytes in culture to investigate the re-lationship between mechanical forces, myofibril assembly, and the localization and translation of (alpha)-myosin heavy chain mRNA. Immunofluorescence and in situ hybridization techniques revealed that contracting myocytes display well aligned myofibrils and a diffuse distribution o...
متن کاملResistance of Cloned 1F5 Chimeric Anti-CD20 Antibody Heavy-Chain Gene to DNA Polymerase due to a Predicted Hairpin Structure
Background: Formation of secondary structure such as DNA hairpins or loops may influence molecular genetics methods and PCR based approaches necessary for genetic engineering, in addition to gene regulation. Materials and Methods: A polymerase chain reaction with splice overlap extension (SOE-PCR) was used to create fully synthetic 1F5 chimeric anti-CD20 heavy- and light-chain genes. The chi...
متن کاملAccumulation of plasma cells in atherosclerotic lesions of Watanabe heritable hyperlipidemic rabbits.
By screening a cDNA library constructed from aortic total RNA derived from Watanabe heritable hyperlipidemic (WHHL) rabbits by differential hybridization, we have obtained a cDNA encoding the kappa light chain of immunoglobulin. Northern blot analysis of total RNA prepared from aortas of WHHL and normal rabbits of various ages revealed that this light-chain mRNA accumulates gradually with age i...
متن کاملMyosin mRNA accumulation and myofibrillogenesis at the myotendinous junction of stretched muscle fibers
Myofiber growth and myofibril assembly at the myotendinous junction (MTJ) of stretch-hypertrophied rabbit skeletal muscle was studied by in situ hybridization, immunofluorescence, and electron microscopy. In situ hybridization identified higher levels of myosin heavy chain (MHC) mRNA at the MTJ of fibers stretched for 4 d. Electron microscopy at the MTJ of these lengthening fibers revealed a la...
متن کاملDistribution of myosin heavy chain mRNA in embryonic muscle tissue visualized by ultrastructural in situ hybridization.
We have localized myosin heavy chain (MHC) mRNAs in cells of intact embryonic chick muscle using high resolution in situ hybridization. Blocks of muscle were aldehyde-fixed prior to detergent treatment and hybridized with a biotinated cDNA probe, followed by colloidal gold-labeled antibodies, before embedment. Labeling was determined to represent MHC mRNA by extensive quantitative comparisons o...
